Role Overview
A Bench Scientist II with a DNA production focus is needed for the Biotherapeutic Expression and Purification group in Cambridge, Massachusetts. This position sits within a busy research laboratory environment that supports non-GMP transient protein lead production for biotherapeutic discovery programs. The selected professional will contribute to DNA production and protein expression workflows that support pre-clinical therapeutic research needs.
The role requires a solid foundation in molecular biology, hands-on experience with nucleic acid production and analysis, and the ability to work on large-scale bacterial culture and DNA production activities using standardized procedures. The scientist will be an important part of the team supporting protein expression and purification efforts.
Key Responsibilities
- Carry out high-volume plasmid DNA production tasks independently, including bacterial growth, plasmid isolation, and downstream sample processing.
- Use and resolve issues with automated lab systems such as Biotage, Opentrons, and similar liquid-handling platforms.
- Run DNA quality checks using gel electrophoresis, nucleic acid measurement, purity analysis, and endotoxin assays.
- Prepare samples for next-generation sequencing and assist with basic review of sequencing outputs.
- Oversee plasmid inventory, sample storage, and tracking using digital databases and sample management tools.
- Keep detailed records in an electronic lab notebook and maintain compliant documentation practices.
- Work closely with expression and purification scientists to ensure DNA is delivered on time for downstream workstreams.
- Support continuous improvement efforts aimed at better throughput, consistency, and turnaround time.
- Share updates on priorities, progress, and potential risks so production goals and deadlines are met.
Requirements
- Bachelor’s degree in Biological Sciences, Molecular Biology, Biotechnology, Biochemistry, or another closely related life-sciences field.
- At least 2 years of experience with molecular biology methods such as plasmid preparation, nucleic acid purification, and quantitative DNA analysis.
- At least 2 years of experience working with bacterial cell culture systems.
- Practical knowledge of DNA quality-control techniques, including gel electrophoresis and sequencing sample preparation.
- Experience using automated or semi-automated laboratory instruments.
- Strong ability to stay organized and juggle several projects in a high-throughput environment.
- Comfort using electronic lab notebooks and sample-tracking systems.
- High attention to detail for repetitive and large-scale tasks.
- Good communication and collaboration skills.
- Strong commitment to lab safety and regulatory compliance.
- Preferred experience includes NGS library preparation or sequencing workflows, Python scripting for data analysis or workflow automation, plate-based 96/384-well workflows, and familiarity with LIMS or sample management platforms such as FreezerPro or Genedata.
